Sequencing service

The VCGS/MCRI sequencing service is your one stop shop for anything 'sequencing' related.

We provide Next Generation Sequencing (NGS) on the Illumina platform as well as Sanger sequencing services for diagnostic analysis and research projects.

We are equipped with a variety of different sequencers - Illumina NovaSeq6000, Illumina NextSeq500, Illumina MiSeq, ABI 3730XL - as well as complementary instrumentation such as 10x Genomics' Chromium controller, Agilent TapeStation, Invitrogen Qubit, Roche LC480 etc.

Our service offers a diverse portfolio of Next Generation Sequencing assays as a full service - this includes sample QCs, library preparation, library QC, and sequencing; but currently excluding bioinformatics.

Current routine assays include:

Whole exome sequencing
Whole genome sequencing – amplification and PCR-free workflows available
RNAseq
Single cell library preparations with the 10X Genomics Chromium system
ChIPseq (starting with your pull-down DNA)
Amplicon based assays (e.g. 16S rRNA)

We also offer custom assay design for your specific research needs on a collaborative basis. Please contact us early during the planning stages of your project, as we can advise on optimal project design.

For all sample submissions:
Please discuss your project with our team before submitting samples for our current assays. We will provide you with a template form to fill in and advise on minimum sample requirements, sample naming, etc.

Samples must be clearly labelled and uniquely identifiable - both within your project and in our laboratory. Samples with identifiers less than four characters will not be accepted.

Examples of acceptable labels include:

123456-1, 123456-2
ABC123-A, ABC123-B
[your initials]-1, [your initials]-2
[your lab acronym]-1, [your lab acronym]-2

Please note, special characters result in downstream issues for data processing and therefore all sample IDs can only contain alphanumeric characters and hyphens.

Service options provided:

Purified DNA pre-mixed with primer: Submission of purified PCR products or plasmid DNA pre-mixed with one primer. The service provided will include running of the BigDye® Terminator v3.1 reaction, BigDye® clean up, as well as capillary separation on our ABI 3730XL instrument.
BigDye® Reactions for Clean Up: Submission of non-purified BigDye® reactions in 10 or 20 ul volume. The service will include BigDye® clean up and capillary separation on our ABI 3730XL instrument.
Cleaned and Dried BigDye Reactions: Submission of purified and dried samples ready for capillary separation.

Sanger sequencing pricing

Batch discounts are available for submission batches of 48+ samples for Purified DNA pre-mixed with primer and BigDye® Reactions for Clean Up.

Submission portal and sample requirements

Our electronic sample submission portal is currently accepting Sanger sequencing samples through LabLink.

For new users: Please request a User ID through LabLink. Once your account has been approved, you will be able to log in and submit samples online.

For existing customers:
Please submit your samples through LabLink

Acceptance criteria for purified DNA pre-mixed with primer samples

Purified DNA and primer submitted in total volume of 15ul with 10pmol total primer (1 ul of 10 uM primer dilution).
If DNA plus primer volume is less than 15 ul, please add nuclease-free water for a total volume of 15ul

Recommended amount of template for purified DNA pre-mixed with primer samples (as determined by spectrophotometric methods)
